Pathogenic for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000051.4(ATM):c.8350G>T (p.Glu2784Ter), citing Ambry Variant Classification Scheme 2023: The p.E2784* pathogenic mutation (also known as c.8350G>T), located in coding exon 56 of the ATM gene, results from a G to T substitution at nucleotide position 8350. This changes the amino acid from a glutamic acid to a stop codon within coding exon 56. This alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation.
