Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000038.6(APC):c.8345_8359dup (p.Tyr2786_Asn2787insThrProPheAsnTyr), citing Ambry Variant Classification Scheme 2023: The c.8345_8359dup15 variant (also known as p.T2782_Y2786dup), located in coding exon 15 of the APC gene, results from an in-frame duplication of 15 nucleotides at positions 8345 to 8359. This results in the insertion of 5 extra amino acids (TPFNY) between codons 2782 and 2786. The amino acid region is well-conserved in available vertebrate species. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
