Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_003000.3(SDHB):c.125T>C (p.Phe42Ser), citing Ambry Variant Classification Scheme 2023: The p.F42S variant (also known as c.125T>C), located in coding exon 2 of the SDHB gene, results from a T to C substitution at nucleotide position 125. The phenylalanine at codon 42 is replaced by serine, an amino acid with highly dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
