Uncertain significance — the classification assigned by Ambry Genetics to NM_006506.5(RASA2):c.829A>T (p.Asn277Tyr), citing Ambry Variant Classification Scheme 2023: The p.N277Y variant (also known as c.829A>T), located in coding exon 9 of the RASA2 gene, results from an A to T substitution at nucleotide position 829. The asparagine at codon 277 is replaced by tyrosine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
