NM_005751.5(AKAP9):c.8235G>T (p.Glu2745Asp) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.E2745D variant (also known as c.8235G>T), located in coding exon 33 of the AKAP9 gene, results from a G to T substitution at nucleotide position 8235. The glutamic acid at codon 2745 is replaced by aspartic acid, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
