NM_002907.4(RECQL):c.820T>G (p.Cys274Gly) was classified as Uncertain significance by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.C274G variant (also known as c.820T>G), located in coding exon 6 of the RECQL gene, results from a T to G substitution at nucleotide position 820. The cysteine at codon 274 is replaced by glycine, an amino acid with highly dissimilar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
