Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_001148.6(ANK2):c.10223G>T (p.Arg3408Leu), citing Ambry Variant Classification Scheme 2023: The p.R3408L variant (also known as c.10223G>T), located in coding exon 38 of the ANK2 gene, results from a G to T substitution at nucleotide position 10223. The arginine at codon 3408 is replaced by leucine, an amino acid with dissimilar properties. This exon is expressed solely in brain (Mohler PJ et al. Circulation. 2007;115(4):432-41). This amino acid position is not well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
