Uncertain significance — the classification assigned by Ambry Genetics to NM_004336.5(BUB1):c.81G>T (p.Trp27Cys), citing Ambry Variant Classification Scheme 2023: The p.W27C variant (also known as c.81G>T), located in coding exon 2 of the BUB1 gene, results from a G to T substitution at nucleotide position 81. The tryptophan at codon 27 is replaced by cysteine, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
