NM_000059.4(BRCA2):c.8197G>C (p.Asp2733His) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.D2733H variant (also known as c.8197G>C), located in coding exon 17 of the BRCA2 gene, results from a G to C substitution at nucleotide position 8197. The aspartic acid at codon 2733 is replaced by histidine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
