Uncertain significance for Primary ciliary dyskinesia — the classification assigned by Ambry Genetics to NM_178452.6(DNAAF1):c.818A>G (p.Lys273Arg), citing Ambry Variant Classification Scheme 2023. This variant lies in the DNAAF1 gene (transcript NM_178452.6) at coding-DNA position 818, where A is replaced by G; at the protein level this means replaces lysine at residue 273 with arginine — a missense variant. Submitter rationale: The p.K273R variant (also known as c.818A>G), located in coding exon 6 of the DNAAF1 gene, results from an A to G substitution at nucleotide position 818. The lysine at codon 273 is replaced by arginine, an amino acid with highly similar properties. This variant was not reported in population based cohorts in the following databases: Database of Single Nucleotide Polymorphisms (dbSNP), NHLBI Exome Sequencing Project (ESP), and 1000 Genomes Project. In the ESP, this variant was not observed in 6500 samples (13000 alleles) with coverage at this position. This amino acid position is conserved in all available species, except pika and x. tropicalis . In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this variant remains unclear.
