Uncertain significance for Hereditary pancreatitis — the classification assigned by Ambry Genetics to NM_001868.4(CPA1):c.817G>A (p.Glu273Lys), citing Ambry Variant Classification Scheme 2023: The p.E273K variant (also known as c.817G>A), located in coding exon 8 of the CPA1 gene, results from a G to A substitution at nucleotide position 817. The glutamic acid at codon 273 is replaced by lysine, an amino acid with similar properties. This amino acid position is poorly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
