Uncertain significance — the classification assigned by Ambry Genetics to NM_001365951.3(KIF1B):c.1395G>A (p.Met465Ile), citing Ambry Variant Classification Scheme 2023: The p.M419I variant (also known as c.1257G>A), located in coding exon 12 of the KIF1B gene, results from a G to A substitution at nucleotide position 1257. The methionine at codon 419 is replaced by isoleucine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. The evidence for this gene-disease relationship is limited; therefore, the clinical significance of this alteration is unclear.
