Uncertain significance for Primary ciliary dyskinesia — the classification assigned by Ambry Genetics to NM_001369.3(DNAH5):c.8022G>C (p.Glu2674Asp), citing Ambry Variant Classification Scheme 2023: The p.E2674D variant (also known as c.8022G>C), located in coding exon 49 of the DNAH5 gene, results from a G to C substitution at nucleotide position 8022. The glutamic acid at codon 2674 is replaced by aspartic acid, an amino acid with highly similar properties. This variant was not reported in population based cohorts in the following databases: Database of Single Nucleotide Polymorphisms (dbSNP), NHLBI Exome Sequencing Project (ESP), and 1000 Genomes Project. In the ESP, this variant was not observed in 6503 samples (13006 alleles) with coverage at this position. This amino acid position is conserved in all available species, except sloth. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this variant remains unclear.
