NM_005477.3(HCN4):c.79G>C (p.Asp27His) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.D27H variant (also known as c.79G>C), located in coding exon 1 of the HCN4 gene, results from a G to C substitution at nucleotide position 79. The aspartic acid at codon 27 is replaced by histidine, an amino acid with similar properties. This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
