Uncertain significance — the classification assigned by Ambry Genetics to NM_001040108.2(MLH3):c.79G>A (p.Glu27Lys), citing Ambry Variant Classification Scheme 2023: The p.E27K variant (also known as c.79G>A), located in coding exon 1 of the MLH3 gene, results from a G to A substitution at nucleotide position 79. The glutamic acid at codon 27 is replaced by lysine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
