NM_000393.5(COL5A2):c.797A>T (p.Asp266Val) was classified as Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.D266V variant (also known as c.797A>T), located in coding exon 11 of the COL5A2 gene, results from an A to T substitution at nucleotide position 797. The aspartic acid at codon 266 is replaced by valine, an amino acid with highly dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
