Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_000384.3(APOB):c.12543A>C (p.Glu4181Asp), citing Ambry Variant Classification Scheme 2023: The p.E4181D variant (also known as c.12543A>C), located in coding exon 29 of the APOB gene, results from an A to C substitution at nucleotide position 12543. The glutamic acid at codon 4181 is replaced by aspartic acid, an amino acid with highly similar properties. This amino acid position is not well conserved in available vertebrate species, and aspartic acid is the reference amino acid in other vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
