Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_000890.5(KCNJ5):c.1253C>G (p.Ser418Trp), citing Ambry Variant Classification Scheme 2023: The p.S418W variant (also known as c.1253C>G), located in coding exon 2 of the KCNJ5 gene, results from a C to G substitution at nucleotide position 1253. The serine at codon 418 is replaced by tryptophan, an amino acid with highly dissimilar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
