NM_017802.4(DNAAF5):c.1253A>T (p.Gln418Leu) was classified as Uncertain significance for Primary ciliary dyskinesia by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.Q418L variant (also known as c.1253A>T), located in coding exon 5 of the DNAAF5 gene, results from an A to T substitution at nucleotide position 1253. The glutamine at codon 418 is replaced by leucine, an amino acid with dissimilar properties. This amino acid position is poorly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
