This variant lies in the PTEN gene (transcript NM_000314.4) at 1021 bases upstream of the translation start (5' untranslated region), where C is replaced by T. Submitter rationale: The c.-1021C>T variant is located in the 5' untranslated region (5&rsquo; UTR) of the PTEN gene. This variant results from a C to T substitution 1021 bases upstream from the first translated codon. This variant is located in the promoter region of the PTEN gene, but its potential impact on PTEN regulation has not yet been investigated (Zhou XP et al. Am J Hum Genet. 2003;73:404-411). This variant was not reported in population based cohorts in the following databases: Database of Single Nucleotide Polymorphisms (dbSNP), NHLBI Exome Sequencing Project (ESP), and 1000 Genomes Project. This position was not covered in the ESP. To date, this alteration has been detected with an allele frequency of approximately 0.001% (greater than 150000 alleles tested) in our clinical cohort. This nucleotide position is well conserved in available vertebrate species. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
