Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_020975.6(RET):c.784G>A (p.Val262Met), citing Ambry Variant Classification Scheme 2023. This variant lies in the RET gene (transcript NM_020975.6) at coding-DNA position 784, where G is replaced by A; at the protein level this means replaces valine at residue 262 with methionine — a missense variant. Submitter rationale: The p.V262M variant (also known as c.784G>A), located in coding exon 4 of the RET gene, results from a G to A substitution at nucleotide position 784. The valine at codon 262 is replaced by methionine, an amino acid with highly similar properties. This alteration was identified in 1/1358 non-cancer control individuals and in 0/57 cases, in a study looking at cancer predisposition mutations in patients with cutaneous melanoma and a history of at least two additional non-cutaneous melanoma primary cancers (Pritchard AL et al. PLoS One. 2018 Apr;13:e0194098). This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
