NM_001378454.1(ALMS1):c.7799A>G (p.His2600Arg) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.H2601R variant (also known as c.7802A>G), located in coding exon 10 of the ALMS1 gene, results from an A to G substitution at nucleotide position 7802. The histidine at codon 2601 is replaced by arginine, an amino acid with highly similar properties. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
