Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_005184.4(CALM3):c.77G>A (p.Gly26Asp), citing Ambry Variant Classification Scheme 2023: The p.G26D variant (also known as c.77G>A), located in coding exon 3 of the CALM3 gene, results from a G to A substitution at nucleotide position 77. The glycine at codon 26 is replaced by aspartic acid, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
