Uncertain significance for Hereditary pancreatitis — the classification assigned by Ambry Genetics to NM_001868.4(CPA1):c.774C>A (p.Asp258Glu), citing Ambry Variant Classification Scheme 2023: The p.D258E variant (also known as c.774C>A), located in coding exon 7 of the CPA1 gene, results from a C to A substitution at nucleotide position 774. The aspartic acid at codon 258 is replaced by glutamic acid, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
