NM_021167.5(GATAD1):c.124G>A (p.Gly42Arg) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.G42R variant (also known as c.124G>A), located in coding exon 1 of the GATAD1 gene, results from a G to A substitution at nucleotide position 124. The glycine at codon 42 is replaced by arginine, an amino acid with dissimilar properties. This amino acid position is well conserved in available vertebrate species; however, arginine is the reference amino acid in other vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
