Pathogenic for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_007194.4(CHEK2):c.76delinsCT (p.Thr26fs), citing Ambry Variant Classification Scheme 2023: The c.76delAinsCT pathogenic mutation, located in coding exon 1 of the CHEK2 gene, results from the deletion of one nucleotide and insertion of two nucleotides causing a translational frameshift with a predicted alternate stop codon (p.T26Lfs*51). This alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation.
