NM_001378454.1(ALMS1):c.7681C>G (p.Gln2561Glu) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.Q2562E variant (also known as c.7684C>G), located in coding exon 10 of the ALMS1 gene, results from a C to G substitution at nucleotide position 7684. The glutamine at codon 2562 is replaced by glutamic acid, an amino acid with highly similar properties. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
