NM_001378454.1(ALMS1):c.7642T>C (p.Phe2548Leu) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.F2549L variant (also known as c.7645T>C), located in coding exon 9 of the ALMS1 gene, results from a T to C substitution at nucleotide position 7645. The phenylalanine at codon 2549 is replaced by leucine, an amino acid with highly similar properties. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
