NM_000314.8(PTEN):c.763G>A (p.Val255Ile) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023. This variant lies in the PTEN gene (transcript NM_000314.8) at coding-DNA position 763, where G is replaced by A; at the protein level this means replaces valine at residue 255 with isoleucine — a missense variant. Submitter rationale: The p.V255I variant (also known as c.763G>A), located in coding exon 7 of the PTEN gene, results from a G to A substitution at nucleotide position 763. The valine at codon 255 is replaced by isoleucine, an amino acid with highly similar properties. In a humanized yeast model, lipid phosphatase activity for this variant was inconclusive (Mighell TL et al. Am J Hum Genet, 2018 05;102:943-955). This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
