NM_001277115.2(DNAH11):c.7610C>T (p.Pro2537Leu) was classified as Uncertain significance for Primary ciliary dyskinesia by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.P2537L variant (also known as c.7610C>T), located in coding exon 46 of the DNAH11 gene, results from a C to T substitution at nucleotide position 7610. The proline at codon 2537 is replaced by leucine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
