Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_003073.5(SMARCB1):c.754A>G (p.Ser252Gly), citing Ambry Variant Classification Scheme 2023: The p.S252G variant (also known as c.754A>G), located in coding exon 6 of the SMARCB1 gene, results from an A to G substitution at nucleotide position 754. The serine at codon 252 is replaced by glycine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
