Uncertain significance for Primary ciliary dyskinesia — the classification assigned by Ambry Genetics to NM_018139.3(DNAAF2):c.751A>C (p.Thr251Pro), citing Ambry Variant Classification Scheme 2023: The p.T251P variant (also known as c.751A>C), located in coding exon 1 of the DNAAF2 gene, results from an A to C substitution at nucleotide position 751. The threonine at codon 251 is replaced by proline, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
