This variant lies in the CASQ2 gene (transcript NM_001232.4) at coding-DNA position 749, where G is replaced by A; at the protein level this means replaces arginine at residue 250 with histidine — a missense variant. Submitter rationale: The p.R250H variant (also known as c.749G>A), located in coding exon 7 of the CASQ2 gene, results from a G to A substitution at nucleotide position 749. The arginine at codon 250 is replaced by histidine, an amino acid with highly similar properties. This variant was reported as contained within a gross duplication of CASQ2 in one Brugada syndrome case; however, clinical details were limited (Mates J et al. Eur J Hum Genet, 2018 07;26:1014-1025). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis.
