Uncertain significance — the classification assigned by Ambry Genetics to NM_013266.4(CTNNA3):c.749A>G (p.Asn250Ser), citing Ambry Variant Classification Scheme 2023: The p.N250S variant (also known as c.749A>G), located in coding exon 5 of the CTNNA3 gene, results from an A to G substitution at nucleotide position 749. The asparagine at codon 250 is replaced by serine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
