Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_001365276.2(TNXB):c.7492+1G>A, citing Ambry Variant Classification Scheme 2023: The c.7492+1G>A intronic variant results from a G to A substitution one nucleotide after coding exon 20 of the TNXB gene. Alterations that disrupt the canonical splice site are expected to cause aberrant splicing, resulting in an abnormal protein or a transcript that is subject to nonsense-mediated mRNA decay. The resulting transcript is predicted to be in-frame and is not expected to trigger nonsense-mediated mRNA decay; however, direct evidence is unavailable. This nucleotide position is highly conserved in available vertebrate species. In silico splice site analysis predicts that this alteration will weaken the native splice donor site and could result in the creation or strengthening of a novel splice donor site. The exact functional effect of the missing amino acids is unknown. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
