Uncertain significance for Primary ciliary dyskinesia — the classification assigned by Ambry Genetics to NM_178452.6(DNAAF1):c.742C>A (p.Arg248Ser), citing Ambry Variant Classification Scheme 2023: The p.R248S variant (also known as c.742C>A) is located in coding exon 6 of the DNAAF1 gene. The arginine at codon 248 is replaced by serine, an amino acid with dissimilar properties. This change occurs in the first base pair of coding exon 6. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
