Uncertain significance for Hereditary pancreatitis — the classification assigned by Ambry Genetics to NM_001868.4(CPA1):c.1243C>A (p.Leu415Met), citing Ambry Variant Classification Scheme 2023: The p.L415M variant (also known as c.1243C>A), located in coding exon 10 of the CPA1 gene, results from a C to A substitution at nucleotide position 1243. The leucine at codon 415 is replaced by methionine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
