Uncertain significance for Primary ciliary dyskinesia — the classification assigned by Ambry Genetics to NM_178452.6(DNAAF1):c.1243A>T (p.Thr415Ser), citing Ambry Variant Classification Scheme 2023: The p.T415S variant (also known as c.1243A>T), located in coding exon 8 of the DNAAF1 gene, results from an A to T substitution at nucleotide position 1243. The threonine at codon 415 is replaced by serine, an amino acid with similar properties. This amino acid position is poorly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
