NM_006440.5(TXNRD2):c.73G>T (p.Gly25Trp) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.G25W variant (also known as c.73G>T), located in coding exon 1 of the TXNRD2 gene, results from a G to T substitution at nucleotide position 73. The glycine at codon 25 is replaced by tryptophan, an amino acid with highly dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
