NM_000041.4(APOE):c.739C>G (p.Leu247Val) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.L247V variant (also known as c.739C>G), located in coding exon 3 of the APOE gene, results from a C to G substitution at nucleotide position 739. The leucine at codon 247 is replaced by valine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
