Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_032043.3(BRIP1):c.738A>G (p.Ile246Met), citing Ambry Variant Classification Scheme 2023: The p.I246M variant (also known as c.738A>G), located in coding exon 6 of the BRIP1 gene, results from an A to G substitution at nucleotide position 738. The isoleucine at codon 246 is replaced by methionine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
