NM_016203.4(PRKAG2):c.737T>C (p.Leu246Pro) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.L246P variant (also known as c.737T>C), located in coding exon 5 of the PRKAG2 gene, results from a T to C substitution at nucleotide position 737. The leucine at codon 246 is replaced by proline, an amino acid with similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
