NM_144670.6(A2ML1):c.737A>G (p.Tyr246Cys) was classified as Uncertain significance by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.Y246C variant (also known as c.737A>G), located in coding exon 8 of the A2ML1 gene, results from an A to G substitution at nucleotide position 737. The tyrosine at codon 246 is replaced by cysteine, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
