Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_007194.4(CHEK2):c.736G>T (p.Val246Leu), citing Ambry Variant Classification Scheme 2023: The p.V246L variant (also known as c.736G>T), located in coding exon 5 of the CHEK2 gene, results from a G to T substitution at nucleotide position 736. The valine at codon 246 is replaced by leucine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
