NM_001365276.2(TNXB):c.7325C>T (p.Thr2442Ile) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.T2442I variant (also known as c.7325C>T), located in coding exon 20 of the TNXB gene, results from a C to T substitution at nucleotide position 7325. The threonine at codon 2442 is replaced by isoleucine, an amino acid with similar properties. This amino acid position is poorly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
