Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_022437.3(ABCG8):c.730G>A (p.Asp244Asn), citing Ambry Variant Classification Scheme 2023: The p.D244N variant (also known as c.730G>A), located in coding exon 6 of the ABCG8 gene, results from a G to A substitution at nucleotide position 730. The aspartic acid at codon 244 is replaced by asparagine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
