Uncertain significance — the classification assigned by Ambry Genetics to NM_001386125.1(OBSCN):c.8587C>T (p.His2863Tyr), citing Ambry Variant Classification Scheme 2023: The p.H2434Y variant (also known as c.7300C>T), located in coding exon 27 of the OBSCN gene, results from a C to T substitution at nucleotide position 7300. The histidine at codon 2434 is replaced by tyrosine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
