Uncertain significance for Primary ciliary dyskinesia — the classification assigned by Ambry Genetics to NM_012144.4(DNAI1):c.728A>C (p.Lys243Thr), citing Ambry Variant Classification Scheme 2023: The p.K243T variant (also known as c.728A>C), located in coding exon 9 of the DNAI1 gene, results from an A to C substitution at nucleotide position 728. The lysine at codon 243 is replaced by threonine, an amino acid with similar properties. This variant was not reported in population based cohorts in the following databases: Database of Single Nucleotide Polymorphisms (dbSNP), NHLBI Exome Sequencing Project (ESP), and 1000 Genomes Project. In the ESP, this variant was not observed in 6503 samples (13006 alleles) with coverage at this position. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since clinical data on this variant is limited at this time, its clinical significance is unclear.
