NM_002519.3(NPAT):c.1239G>C (p.Gln413His) was classified as Uncertain significance by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.Q413H variant (also known as c.1239G>C), located in coding exon 13 of the NPAT gene, results from a G to C substitution at nucleotide position 1239. The glutamine at codon 413 is replaced by histidine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
