Uncertain significance for Cardiovascular phenotype; Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_001042492.3(NF1):c.7256T>C (p.Leu2419Pro), citing Ambry Variant Classification Scheme 2023: The p.L2398P variant (also known as c.7193T>C), located in coding exon 48 of the NF1 gene, results from a T to C substitution at nucleotide position 7193. The leucine at codon 2398 is replaced by proline, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
